Arabidopsis lysin motif/F‐box‐containing protein InLYP1 fine‐tunes glycine metabolism by degrading glycine decarboxylase GLDP2

چکیده
Lysin motif (LysM) is a carbohydrate-binding module often found in secreted or transmembrane proteins living organisms from prokaryotes to eukaryotes. Thus far, all characterized LysM-containing plants are plasma membrane-resident receptors co-receptors playing roles plant–microbe interactions. Here, we interrogate the Arabidopsis LysM/F-box-containing protein InLYP1 and reveal its function glycine metabolism. was mainly expressed by vigorously growing tissues, encoding nuclear-cytoplasmic protein. We validated as part of SKP1-CULLIN1-F-box E3 complex for mediating degradation. The decarboxylase P-protein 1 (GLDP1) identified an InLYP1-interacting both immunoprecipitation/mass spectrometry yeast two-hybrid library screening. could also interact with GLDP2, paralog GLDP1 weaker catalytic activity, mediate degradation GLDP2 but not GLDP1. Interestingly, GLDPs be O-glycosylated form homodimers heterodimers. Overexpression InLYP1L9A dominant-negative variant cause seedling germination retardation on medium containing glycine. Collectively, these results shed light plant intracellular proteins, suggest that may deplete facilitate decarboxylation Arabidopsis.

T-protein of glycine decarboxylase from Solanum tuberosum.
The Gly decarboxylase multienzyme complex (Gly cleavage system, EC 2.1.2.10) catalyzes the conversion of Gly into COz, NH3, and 5,10-methylene-tetrahydrofolate, reducing NAD+ to NADH. The enzyme has been isolated from animals, plants, and bacteria-in animals and plants it occurs exclusively in the mitochondrial matrix, and in photosynthetic tissue it can be found in very large amounts (Bourguig...

GDC is the key enzyme of Gly catabolism in bacteria, animals, and plants. In plants this mitochondrial multienzyme complex is involved in the photorespiratory carbon oxidation cycle. Together with Ser hydroxymethyltransferase, it catalyzes the conversion of two molecules of Gly into Ser, COz, and NH3, accompanied by the reduction of NAD+ to NADH (Sarojini and Oliver, 1983). The enzyme complex c...
متن کاملThe glycine decarboxylase system: a fascinating complex.
The mitochondrial glycine decarboxylase multienzyme system, connected to serine hydroxymethyltransferase through a soluble pool of tetrahydrofolate, consists of four different component enzymes, the P-, H-, T- and L-proteins. In a multi-step reaction, it catalyses the rapid destruction of glycine molecules flooding out of the peroxisomes during the course of photorespiration. In green leaves, t...
